The o-ring around the regulator needs a smooth surface within the reservoir to get a good seal. Any marks going lengthways across where the seal is sitting will cause a leak. DEET doesnt really repel ticks.
However Permethrin does a good job as ticks are repelled or killed. It can be mixed into a solution for treating clothing. Or, some dog bedding flea sprays contain Permethrin.

Usual practice for an empty fill is to cock the rifle, so the hammer isnt pressing against the valve stem.
If it still leaks, remove the valve and check its clean and the face of valve and valve body are not scratched
